چکیده

pposites really do attract. Results on page 101 by Oliveria et al. show that, although they perform opposing functions, a kinase and a phosphatase linked to synaptic plasticity are close neighbors at the plasma membrane. The two are brought together by the scaffolding protein AKAP79 into a complex that may be important for efficient learning. The close proximity of the three proteins is uncovered by a powerful microscopy technique, fluorescence resonance energy transfer (FRET). Using this technique in COS cells, the group shows that protein kinase A (PKA) and the phosphatase calcineurin (CaN) bind to sites on membrane-targeted AKAP79 that are spaced only nanometers apart. In neurons, AKAP79 is known to O

Bioinspired optofluidic FRET lasers via DNA scaffolds.

Optofluidic dye lasers hold great promise for adaptive photonic devices, compact and wavelength-tunable light sources, and micro total analysis systems. To date, however, nearly all those lasers are directly excited by tuning the pump laser into the gain medium absorption band. Here we demonstrate bioinspired optofluidic dye lasers excited by FRET, in which the donor-acceptor distance, ratio, a...

Crosslinking of cell-derived 3D scaffolds up-regulates the stretching and unfolding of new extracellular matrix assembled by reseeded cells.

Elevated levels of tissue crosslinking are associated with numerous diseases (cancer stroma, organ fibrosis), and also eliminate the otherwise remarkable clinical successes of tissue-derived scaffolds, instead eliciting a foreign body reaction. Nevertheless, it is not well understood how the initial physical and biochemical properties of cellular microenvironments, stem cell niches, or of 3D ti...

Assembling programmable FRET-based photonic networks using designer DNA scaffolds

DNA demonstrates a remarkable capacity for creating designer nanostructures and devices. A growing number of these structures utilize Förster resonance energy transfer (FRET) as part of the device's functionality, readout or characterization, and, as device sophistication increases so do the concomitant FRET requirements. Here we create multi-dye FRET cascades and assess how well DNA can marsha...

Highly Sensitive FRET-Based Fluorescence Immunoassay for Detecting of Aflatoxin B1 Using Magnetic/Silica Core-Shell as a Signal Intensifier

Background: Recently, some new nanobiosensors using different nanoparticles or microarray systems for detection of mycotoxins have been designed . However, rapid, sensitive and early detection of aflatoxicosis would be very helpful to distinguish high-risk persons. Objectives: We report a highly sensitive competitive immunoassay using magnetic/silica core shell as a signal intensifier for the d...

Fluorescence characterization of immobilization induced enzyme aggregation.

Förster resonance energy transfer (FRET) has been used to show that the average distance between proteins decreases when the protein is immobilized within polymer scaffolds, indicating that the immobilization process is inducing aggregation.
